张纪岩,军事科学院军事医学研究院教授、博士生导师,国家杰出青年科学基金获得者。中国免疫学会理事,《中国免疫学杂志》副主编。2007年第四届中国青年女科学家奖获得者、2008年被评为总后勤部科技新星2012年入选军队高层次科技创新拔尖人才。主要从事炎症消退与免疫信号调控研究,共发表SCI论文47篇。其中,在Mol Cell, Cancer Res, Mol Cell Biol等杂志发表第一或共同第一作者SCI论文9篇;在Cell Rep, Hepatology, PNAS, Oncogene, Cell Death Differ, Mol Biol Cell, Mol Ther, J Immunol等杂志发表独立或合作通讯作者SCI论文24篇。参与的工作发表在Cell, Nat Immunol等杂志上。

内容简介

Neddylation has emerged as a potential regulator of T cell activities to TCR engagement, but its contribution to T cell-mediated immunity to blood-stage malaria remains unclear. Here we demonstrate activation of neddylation pathway in T cells during Plasmodium yoelii 17XNL infection, which is essential to guarantee parasite control and host survival. Mechanistically, we show that apart from promoting CD4+ T cell activation, proliferation, and development of protective Th1 response as suggested previously, neddylation is also required for supporting CD4+ T cell survival, mainly through Bcl-2 mediated suppression of the mitochondria-dependent apoptosis. Furthermore, we provide evidence that neddylation contributes to follicular helper T cell differentiation, probably via augmenting Itch targeted FoxO1 degradation, thereby facilitating germinal center formation and parasite-specific antibody production. This study identifies neddylation as a positive regulator of anti-Plasmodium immunity and provides insight into an involvement of such pathway in host resistance to infectious diseases
